The following independent study explores the biological functions of microRNAs—their role as the master regulators of gene expression and their relevance to thyroid cancer biology and processes. While this study includes microRNA assessed by the ThyraMIR®v2 assay, it should not be interpreted as a validation study for ThyraMIRv2 or the ThyGeNEXT® + ThyraMIRv2 combination testing platform.

Xiang D, et al. mir-222 expression is correlated with the ATA risk stratifications in papillary thyroid carcinomas. Medicine. 2019:98(25):e16050.

This study analyzed miR-222 expression, one of the most consistently overexpressed microRNAs found in papillary thyroid carcinoma (PTC), and its correlation with American Thyroid Association® (ATA) risk stratifications. Researchers analyzed FFPE tissue samples from 100 patients (90 with PTC and 10 with multinodular goiter) using RT-PCR detection methodology to measure the expression of individual miRNAs.

The findings revealed that miR-222 levels were significantly higher in PTC tissues compared to benign goiter tissues. miR-222 expression correlated with aggressive clinicopathological features such as larger tumor size, capsular and vascular invasion, and lymph nodes metastasis. Within the ATA risk-stratification groups, miR-222 was most highly expressed in the high-risk group and least expressed in the low-risk group. However, miR-222 showed no correlation with tumor-node-metastasis (TNM) staging in this study.

The data provide further support of the value that microRNAs can bring in the risk-stratification of indeterminate thyroid nodules (ITN), providing deeper insights to help guide clinical decisions in the management of patients with PTC.

Patient management decisions are based on the independent medical judgment of the physician and molecular test results should be taken into consideration in conjunction with all relevant imaging, clinical findings, patient and family history, as well as patient preference.
